英文摘要:       Technique products of polybrominated diphenyl ethers (PBDEs) were widely used as flame retardants  and have been banned due  to their persistence and higher  toxicity since 2009.  However, decabromodiphenyl ether  is still utilized  in China due  tolower toxicity. In the environment, higher brominated PBDEs could be transformed  to lower brominated diphenyl ethers  through biotic or abiotic  processes, potentially producing higher toxicity.  Thus, study on  the behavior  of lower brominated  PBDEs, especially their metabolism, is very important.
     The  analytical method  of  PBDEs  mainly referenced  the  standard protocol  for polychlorinated biphenyls (PCBs)  analysis. Due to the potential  influences caused by sample pretreatment, matrix effect and instrumental factors in the analytical procedure,this   thesis   firstly  focused   on   the   evaluation   method   for   those  influences   in environmental analysis. The method was successfully applied in the evaluation of those influences on  the BDE-99  analysis. The  solution to avoid  those influences  was also proposed, using molecular ions instead of fragmental ions as the quantify ions.
     The   metabolism   process   of    PBDEs   mainly   includes   methoxylation   and hydroxylation. The mono-  substitute compounds for OH- and  MeO-PBDEs have 837 isomers, respectively. However, there are only over 40 commercial standards for each. It is impractical to synthesize all of them for qualitative analysis. This thesis established a regression model with dummy variables based on the behaviors of MeO-PBDEs and PBDEs on GC-MS for the qualitative of MeO-PBDEs. The newly developed regression model was successfully used to predict the structures of unknown MeO-PBDEs.
     A  bioaccumulation  directed   screening  method  was   developed  for  unknown metabolites of PBDEs. Retention  times and bromide precursor ion scan  mode on LC-MS/MS were  combined to  screen potential  contaminants in  environmental samples.Two unknown compounds of interest in real soil samples were screened as the potential bioaccumulative brominated contaminants for terrestrial ecosystems.
    Finally,  both   in  vitro  and   in  vivo   exposure  experiments  of   BDE-28  were performing using  pumpkins.  The results  showed that  MeO-PBDEs  and OH-PBDEs metabolites existed in pumpkin tissues. The structures of three unknown MeO-PBDEs were    indentified.    The    OH-PBDEs    metabolites    were   bioaccumulative.    The biotransformation  of   PBDEs  to   para-   substituted  MeO-PBDEs   is  an   important metabolism pathway in pumpkins.          
          In summary, this thesis developed analytical methods for PBDEs and their metabolites which were successfully applied in metabolism study of BDE-28 in  pumpkin plant. Those methods could not only be used for the envrionmental fate studies on PBDEs, but also on other pollutants.
